低温乳制品中金黄色葡萄球菌实时荧光PCR检测方法的建立和应用

Abstract: In this paper,a real-time fluorescent Polymerase Chain Reaction (PCR) rapid detection method system for Staphylococcus aureus was established fortherapid release of low-temperature dairy products.It can complete enrichment and rapid detection within 24h Moreover, internal standard gene amplification was added in thereal-time fluorescent PCR reaction to monitor whether the PCR amplification reaction was inhibited.Specificity, sensitivityand consistency with themethodof National Standard for Food Safety-Microbiological Examination of Food-Detection of Staphylococcus aureus (GB 4789.10—2016) were validated according to the requirement of ISO 16140-2:2016,and sample application tests are carried out in dairy factories. the results showed that the method has good specificity,the detection limit is1CFU/25g(mL),Theresultsof artificial contaminationsample andactual sample testing showed thatthe method was in good consistency with GB 4789.10—2016 in several low-temperature dairy products, with a relative accuracy of 100% .In addition,when the method results were negative,the internal control results were normal, which played a role in monitoring the reaction process.In summary,the method in this study can be used for the rapid qualitative detection of Staphylococcus aureus in low-temperature dairy products.
