Determination of Aflatoxin B1 in CerealsbyHPLC with Post-Column Derivatization

Abstract:This article establishesa rapidand eficient high-performance liquid chromatography method for the determination ofaflatoxinI β1 in cereals.Samples were extracted with a methanol and water mixture at a volume ratio of 70:30 and then the extract was purified byaspecific immunoaffinity column for aflatoxin B -1 .In the elution step,methanol and water at a volume ratio of 1:1 were usedas the eluent,followed by detection with afluorescence detector combined with photochemical post-column derivatization. The results show that aflatoxin B1 has a good linear relationship in the concentration range of (20 0.1~40.0ng/mL with thecorrelation coefficient reaching O.999 6.The average recoveries of the method are inthe range of 92.4% 100% ,and the relative standard deviationsare between 2.0% ( 2.7% . The limit of quantification is determined to be 0.068μg/kg .All performance indicators meet the relevant detection standards.
