Abstract:Gelatin methacrylate (GelMA)was prepared using gelatin and methacrylic anhydride.Different mass concentrations of polyethylene glycoldiacrylate(PEGDA)wereadded,followed bymixingand photocrossinking topreparegelatinmethacrylate-polyethylene glycoldiacrylatecompositehydrogels(GelMA-PEGDAhydrogels)with diferentstifneses.These hydrogels were used to bionicall construct 3D breast cancer models of extracelular matrices with different stifnesses,soastoinvestigatetheeffectsofextracellularmatrices withdiferentstiffnessesonthegrowthofbreastcancer cells MDA - MB -231 and MCF -7 and their sensitivity to chemotherapeutic drugs paclitaxel and doxorubicin.Nuclear magneticresonancespectroscopycharacterizationshowed thatthegrafting rateof gelatinand methacrylicanhydridewas (2号 84.38% ;Fourier transform infrared spectroscopy characterization confirmed that GelMA was mixed with diffrent mass concentrationsofPEGDAand then photocrosslinked to form(5G1P,5G3P)hydrogel crossinks.Dynamic thermomechanical analyzerwas used to characterize the stiffess of hydrogels.The weighing method was used to characterize hydrogel swelling,and the massloss method was used to characterize the hydrogel degradationrate.Scanning electron microscopy wasusedtocharacterizethemorphologyofhydrogels.Live/dead staining wasusedtocharacterizethecytocompatibilityof hydrogels ,and the CCK -8 assay was used to evaluate the sensitivity to chemotherapeutic drugs.
