Determination of Three Hyoscypane Alkaloids in Medicinal Wine by UPLC-MS/MS Method Based on Carbon S Technology

ZHONG Xue,MENG Chunyang, WU Yutian, ZHOU Yibing,LIN Ye, ZOU Lu* (Guizhou Provincial Center for Disease Control and Prevention, Guiyang 55ooo4, China) Abstract: Objective: To establish a rapid method for the detection of three types of hyoscypane alkaloids in medicinal wine.Method: The samples were extracted with acetonitrile and purified with Carbon S adsorbent.The mobile phase was methanol-5 mmol L-1 ammonium formate (containing 0.1% formic acid),and the detection was caried out in the multi-reaction monitoring positive ion mode.Result: The three kinds of hyoscypane alkaloids had a good linear relationship in the range of 2.0μg⋅L-1 to 100.0μg⋅L-1, and the correlation coefficient was greater than 0.999; the detection limit was 0.10μg⋅L-1 to 0.25μg⋅L-1 ,and the quantification limit was 0.25μg⋅L-1 to 1.00μg⋅L-1 the recovery rate of standard addition was 70.23% to 90.10% ,and the relative standard deviation was 1.71% to 7.44% Conclusion: This method is simple and accurate,and can be used for the rapid analysis of hyoscypane alkaloids in medicinal wine.
