AbstractObjective:To establishamouse modelofslep disorders bysimulatingconditions inaconfined environment.Methods:A total of 4O KM male mice were randomly divided into four groups( n=10 each):control group(CON group),light change group(Lgroup),restraint group(R group)and light change plus restraint group(LR group).After30days of intervention,mice were subjectedtoasodiumpentobarbital-inducedsleeptest,inwhichsleep latencyandsleepdurationwererecorded.Hypothalamic tissesandserumsampleswere takenforthe measurementof slep-related neurotransmitersorhormones.The enzyme-linked immunosorbent assay(ELISA)was used to detect γ -aminobutyric acid(GABA)dopamine(DA),and 5- hydroxytryptophan(5-HT)inthehypothalamustisues.Serumhormones,includingorexinA(OXA),corticotropin(CO)corcotropin-releasing hormone(CRH)andadrenocorticotropic hormone(ACTH)werealso measured toevaluate themodeling ffects underdiffrentconditions.Results:ThebodyweightsoftheRandLRgroupsattheendof theexperimentweresignificantlylower ( P<0.05 )compared withthe CON group. Under light change plus restraint condition,sleep latency was significantly prolonged ( P<0.05 ) and sleep duration was significantly shortened( P<0.01 ) in mice. In the measurement of hypothalamus-related neurotransmitercontents,comparedwith theCONgroup,the mice intheLR grouphadsignificantlylowerGABAand 5-HTlevels ( P<0.05 )and higherDA levels( P<0.05, );in the measurement of serum-related hormone contents,compared with the CON group,the LR group had significantly higher OXA,CORT and ACTH levels( P<0.05 for each)and CRH levels did not change significantly( P>0.05 ).Conclusion:Light plus restraint treatment in a confined environment can affect the levels of relevant neurotransmitersandhormones inthe hypothalamusandserum,disruptnormalsleepprocess inmice,andestablishamodelof sleep disorders.
