肝郁脾虚方对失眠大鼠下丘脑 γ -氨基丁酸 和谷氨酸含量的影响

AbstractObjective:This study aims to investigate the eects of Ganyu Pixu Formula(GYPXF) on γ -aminobutyric acid(GABA)and Glutamate(Glu)levelsinthehypothalamusof insomniarats.Methods:Thirty-twospecificpathogen-fre(SPF)-grade male Sprague Dawley(SD)rats wererandomly divided into four groups,with 8rats ineach group:normal control group,model controlgroup,GYPXFgroup,anddazepamgroup.Exceptforthoseinthenoralcontrolgroup,allratsweresubjectedtotheodelingforliverdepresio-speedeficncyisomiasdromebhonictail-capngstiulationintraperitonealp-loylalanine(PCPA)injections andirregular feeding pattems(fastingonodd-numbered days withad libitum wateraccess).After7 daysof intragastricadministrationwithcorresponding treatments,hypothalamicGABAandGlulevelswerequantifiedusingenzyme-linked immunosorbent assay(ELISA),with subsequentcalculationof Glu/GABAratios.Results:Compared with thenormal controlgroup,rats inthemodel groupexhibitedmanifestationsoflethargyandcoarse,lustreless fur,andsignificantlydecreased body weight( P<0.01). The hypothalamic GABA was significantly lower( P<0.05) ,while Glu level was significantly higher in the model group than the normal control group( P<0.01 ),resulting in a significantly higher Glu/GABA ratio ( P<0.01 ). In comparison withthemodel group,ratsinboth theGYPXFgroupanddiazepam groupdemonstratedclinicalimprovementscharacterized by enhanced mental status and restored fur luster,along with significantly increased body weight( P<0.05 ). Specifically, rats in the GYPXF group exhibited significantly elevated GABA content( P<0.05 ),significantly reduced Glu levels in hypothala1 nus(P<0.01) ),and significantly decreased Glu/GABA ratio( P<0.01 ). Rats in the Diazepam group showed more pronounced neurochemical alterations with significantly increased GABA content ( P<0.01 ).Conclusion:GYPXF effectively ameliorates sleepdisordersinatswithliverdepresion-spleendeficiencyisomniaotentiallythroughadualmodulatioofneurotraier balance-upregulatingGABAsynthesiswhiledownregulatingGluproduction,therebyrectifying thepathologicalGlu/GABAimbalance in hypothalamic regulatory circuits.
