Study on the Quantitative Method of Droplet Digital PCR for Bovine-DerivedMaterials in MeatProducts

Abstract: Objective: To address the problem of meat adulteration, a highly sensitive quantitative detection method for bovine-derived materials was established to distinguish between unintentional contamination and intentional adulteration,and to provide technical support for food safetysupervision.Method: Fresh beef was tested by droplet digital PCR using bovine-specific primers and probes,and a conversion model between beef quality and nucleicacid copy number was established.The specificity,accuracyandanti-interferenceabilityofthe method were verified by testing negative samples (chicken,mutton,pork),mixed meat samples and commercial bef products. At the same time,the feasibility of directly establishing a linear regression equation between beef quality and nucleic acid copy number for quantitative detection of bovine-derived materials was explored. Result: There was a significant linear relationship between beef quality and DNA content ( R2=0.998 7 ) and between DNA content and copy number ( R2=0.9968 ). Based on this, the conversion formula between beef quality and copy number (ν= 0.037 6x-0.516 5 )was derived,which can be used for the quantitative detection of bovine-derived materials.The method has good specificity,accuracy and strong anti-interference ability. The linear equation of beef quality and copy number (y=35.841x-11.523, R2=0.997 3 ) was directly established, which is suitable for quantitative detection of bovine-derived materials in meat products,simplifes the detection process,and improves the detection eficiency. Conclusion: Droplet digital PCR technology can accurately quantify bovine-derived materials in meat products with high accuracyand stronganti-interference ability,and is suitable for food adulteration identification and daily monitoring work of regulatory authorities.
