Study on Purification Process of Platycodon grandiflorum Polysaccharide by Macroporous Resin and Its Biological Activity

Abstract: In this study,Platycodon grandiflorum was used as theraw material to extract crude polysaccharides from Platycodon grandiflorum by the water extraction and alcohol precipitation method. By comparing the purification effects of four diferent types of macroporous resins,namely AB-8,NKA-9,D1o1,and D3520,on Platycodon grandiflorum polysaccharides,the best resin was screened.The results show that the best resin screened outisAB-8,with a decolorizationrate of 93.82% ,aprotein removal rate of 90.74% ,andapolysaccharideretention rate of 79.88% .The Oxford Cup method antibacterial experiment indicated that both the polysaccharides of Platycodon grandiflorum before and afterpurification hadcertainantibacterial efectson Pseudomonas aeruginosa andEscherichia coli,andthedifference inantibacterial abilitybetweenthe two was relativelysmall.Theantioxidant activity of Platycodon grandiflorum polysaccharides was determined by measuring the hydroxyl radical scavenging rate.The results showed that at the same concentration,the purified Platycodon grandiflorum polysaccharides had stronger antioxidant activity.The research results can provide useful references for the development andutilizationof Platycodon grandiflorum polysaccharides.
