效液相色谱法测定皱皮木瓜成熟果实中维生素C

Abstract: A high performance liquid chromatography method was established to determine vitamin C of Chaenomeles speciosa. stationary phase was Inertsil NH2 C18 250mm×4.6mm , 5μm ). The mobile phase was acetonitril- 0.1% oxalic acid solution (85:15) . The flow rate was 1.0mL⋅min-1 , the column temperature was 30°C , and the detection wavelength was 424nm . Taking Chaenomeles speciosa speciosa as the determination object, oxalic acid solution as the extraction solvent, the mass concentration and ultrasonic time were investigated. The results showed that the extraction efficiency was the highest when 0.5% oxalic acid solution was used as the extraction solvent and ultrasonic time was 20min . The vitamin C showed a good linear relationship with the range between 4.27μg⋅mL-1 to 21.36μg⋅mL-1 , the correlation coefficient was 0.999 6. The recoveries were 97.80% to 99.04% , the relative standard deviation were 0.50% to 0.83% . This method is simple and accuracy is high, it can be used for quality control and evaluation of mature fruit in Chaenomeles speciosa.
