Study on the Determination of Aflatoxins B Group and G Group in Foods by Automated Solid-Phase Extraction-High Performance Liquid Chromatography

Abstract: To achieve precise detection of aflatoxins B group (AFT B1: AFTB2 )and G group (AFT G1 AFT G2 ) incorm,rice,andpeanutoil,thisstudyreferredtothethirdmethodinNationalFoodSafetyStandardDetermination of Aflatoxins B and G Group in Food (GB 5009.22—2016) and established a fully automated solid-phase extractionhigh-performance liquid chromatography detection method. Samples were extracted with 70% methanol in water, centrifuged, diluted with PBS buffer containing 0.1% Tween-20, purified through an aflatoxin immunoaffinity column,eluted with methanol,andconcentratedunderanitrogen stream.Isocratic elution was performedona Waters BEH- C18 column, and detection was carried out by a fluorescence detector (excitation wavelength 365nm ,emission wavelength 463nm ).The results showed that the target analytes exhibited good linearity in the mass concentration range of 0.5μg⋅L-1 to 100.0μg⋅L-1 ,with recoveries of 83.3% to 103.6% at spiking levels of 0.5, 2.0, 5.0μg⋅kg-1 and relative standard deviations below 5% .This method offers high throughput, high automation, and good accuracy and repeatability, meeting the routine detection needs of aflatoxins in grains and oils.

黄曲霉毒素是由黄曲霉菌和寄生曲霉菌产生的一类强致癌性真菌毒素,其中B族(AFT ΔB1 、AFTB2 )与G族(AFT G1 、AFT G2 )在粮油食品污染中最为常见,对人体健康构成严重威胁。(剩余3772字)
