Abstract: Objective: To establish a method for the determination of aloin content in health products using high performance liquid chromatography. Method: Samples were ultrasonically extracted with 55% methanol aqueous solutionby volume.Methanol- 0.1% phosphoric acid by volume was used as the mobile phase,and separation was performed on a CNW C18 column ( (250mm×4.6m m, 5.0μm )under isocratic elution. Quantification was conducted using a standard calibration curve. Result: A good linear relationship was observed for aloin concentrations ranging from 0mg⋅L-1 to 50mg.L-1 ,with a correlation coefficient (R2) of 0.999 7. The method detection limit was 3mg⋅kg-1 to 15mg⋅kg-1 ,and the quantitation limit was 10mg⋅kg-1 to 50mg⋅kg-1 .For both solid and liquid samples,spiked recovery experiments were performed at 1,2,and 5 times the respectivequantitation limits,yielding recoveriesof 87.8%~99.7% and relative standard deviations of 2.11%~5.08% .Conclusion: The established method offers high recovery and good stability, making it suitable for qualitative and quantitative analysis of aloin in various types of health products.
