基于荧光光谱技术的甘蔗多酚快速检测方法

Abstract: As an important natural antioxidant substance, the rapid and accurate detection of sugarcane polyphenols is of great significance for foodqualityevaluation.In this study,arapid detectionmethod forsugarcane polyphenols based on fluorescence spectroscopy technology was established.Single-factor experiment combined with Box-Behnken response surface design were used to optimize the sample pretreatment conditions and fluorescence detection parameters. The optimal extraction conditions were determined to be methanol volume fraction of 80% , solid-liquid ratio Ξ(g:mL) of 1:5 ,ultrasonic extraction for 35min ,and ultrasonic powerof 400W. Theoptimal detection conditionswere an excitation wavelength of 278nm. ,an emission slit width of 5nm ,andaphotomultiplier voltage of 620V. The precision test shows that the relative standard deviations within a day, between days and among personnel were all no more than 3.6% ,and the recovery rate of standard addition was 98.3% to 101.7% .15actual sugarcane samples showed that the fluorescence method had good agreement with the Folin-Ciocalteu colorimetric method,butitsdetection time isrelativelyshort,only2.5minutes.Themethod issimple,rapidandaccurate,and provides an effective technical means for the quality control and nutritional evaluation of sugarcane products.
