高效液相色谱-串联质谱法测定灵芝中3种麻黄碱含量

Abstract: An analytic method was developedto investigate ephedrine hydrochloride,pseudoephedrine hydrochloride and methyl ephedrine hydrochloride in Ganoderma lucidum by high performance liquid chromatography-tandem massspectrometry.The analyteswere separated by gradient elution using 0.1% (v/v) )formic acid aqueoussolution and 1 0.1% formic acid methanol solution as mobile phases.The analytes were extracted with 0.1% (v/v) formic acid aqueous solution and -0.1% (v/v) formic acid methanol solution (95:5). The external standardization method was used forthe quantitative analysis.Thecalibration curves of the 3 target analytes were linearin the massconcentration range of 0~10ng/mL ,with correlation coefficients greater than 0.999.The method detection limit range and the method quantification limit range were 2.0~6.0μg/kg and 6.4~19.6μg/kg ,respectively.The average spiked recoveries for the 3 target analytes at three levels ranged from 97.3% to 112.0% ,and the relative standard deviations(RSDs) were less than 10% .The method is simple with highly sensitive,and can be used for theanalysis anddetermination of ephedrine hydrochloride,pseudoephedrine hydrochloride and methyl ephedrine hydrochloride in Ganoderma lucidum.
