Determination of Three Chemical Components in Rapeseed Pollen

Abstract: Objective: To establish a high performance liquid chromatography (HPLC) method for the simultaneous determination of rutin,quercetin and kasanferol contents inrapeseed polln.Method: An Agilent Poroshell 120EC-C18 chromatographic column was used, with a gradient elution using methanol- .0.4% phosphoric acid as the mobile phase. The flow rate was 1mL⋅min-1 ,the injection volume was 5μL. ,and the column temperature was 30∘C . The detection wavelength is 368nm .Result: The correlation coefficients of rutin, quercetin, and kaempferol were all greater than 0.999 within their respective concentration ranges,indicating a good linear relationship.The relative standard deviations (RSD) of the three substances in the precision experiment were 0.62% 0.65% ,and 0.51% 0 respectively. In the stability experiment, the RSD of the three substances were 3.26% 0 2.70% and 1.86% ,respectively. In the repeatability experiment, the RSD values of the three substances were 1.29% 1.85% ,and 2.85% ,respectively. The average recovery rates were 100.44% 99.34% ,and 99.65% ,respectively. Conclusion: This determination method is easy tooperate,has high precision,good stability,accurateandreproducible detection results,andcanbeused for the determination of rutin, quercetin and kaempferol content in rapeseed pollen.
