Analysis of HMW-GS composition of two Aegilops diploid species based on relative mobility

Abstract:Aegilopsisconsideredasanimportantgenesourceforimprovingmodercommonheat,whichhasmanyexcelentgenesor traits.Inrdertounderstandthegeneticdiversityofhighmolecularweightgluteninsubunits(HMW-GS)inAe.diploidspecies,odiumdodecylsulfate polyacrylamide gelelectrophoresis(SDS-PAGE)was usedtoanalyze HMW-GS in40Ae.acession(Ae.comosa andAe.markgrafi).Theresultsshowedthetotalof17elativemobilitieswereidentifiedamong the40acessions,ofichfiveelativemobilities(0.82~0.98)werelowerthanthatofChineseSpring1Dx2(1.0O),andonerelativemobilty(1.49)washigherhanthat of ChineseSpring1Dy12(1.40).Analysisof geneticsimilaritycoeficients(GS)among theaccessionsrevealedarangeof0.00~00, withanaveragevueofO3.And,itasfoundtatteverageG(.58)ofAe.markgraficessosassiificantlygran that of Ae. comosa accessions(O.31). The clustering results showed that 40Ae . accessions were divided into two groups at the GS 0.10, of which19Ae.comosaacessonswerecusteredinGroup1.And,theGroup2inluding21Ae.ccessonscouldbefurtherdividedinto two sub-groups,which indicated that Ae.accessions clustering together had the same species.
