Cloning and bioinformatic analysis of NHXl gene in Shenqiuhong Hippophae rhamnoides

Abstract:UsingShenqiuhongHippophaehamnoidesastheexperimentalmaterial,thegeneencodingtheNHX1proteiof heNa+/H+ antiporter(NHX)familywascloned,andbioinformaticanalysis was performedonthe HippophaerhamnoidesNHX1protein(HrNHX1).Theresults indicatedthatthefullengthofthe HrNHX1 gene wasapproximately6OObp,encodinganacidic hydrophobicpro tein consisting of 538 amino acid residues.The molecular formula of this protein was C2754H4269N672O759S21 ,with a total of 8 502 atoms andarelativemolecular massof 59637.71.HrNHX1belonged toastableprotein,containing13transmembrane domains,ackinga signalpeptid,beingano-secretoryproteinndcontainingoneNa+/HexchangerdomaincharacteristicoftheNHXproteinfamily. Subcelularlocalizationanalysisshowedthatthe HNHX1proteinwaslocalizedonthevacuolarmembrane.Homologyalignmetanalysis indicated thattheHrNHX1proteinsequencehadhighsequencesimilaritywiththeNHX1proteinsequencesofGosypiumaimondi,Gossypiumhirsutum,Morusnotabilis,ndHeveabrasiliensis.PhylogenetictreeanalysisfoundthattheHrHX1proteinsequence clusteredonthesameevolutionarybranchwiththeNHXproteinsequencesofriceandwheat,whichhaddroughtresistanceandsalt tolerancefunctions,sugestingtheyhadacloseevolutionaryrelationship.The HrNHX1geneplayedanimportantroleinenhancing plantdroughtresistanceandsalttolerance,andregulatingtheactivityofitsencodedHrNHX1proteinorthexpresionlevelofthis gene could effectively improve plant adaptability to drought and salt stress.
