Abstract:Thealternativeoxidase(AOX)familygeneinAmorphophaluskonjacK.Kochwasanalyzedatthewhole-genomelevel.The resultsshowedthat therewerefouralternativeoxidasefamilygene membersinAmorphophalushonjac,namedAkAOX1toAAOX4. Their proteinlengthrangedfrom330to361aa,molecularweightsfrom37087.52to41243.83u,nd isoelectricpointsfrom5.97to 7.27.Theywereprimarilylocated inthecytoplasmandchloroplast.TheAmorphophalus honjacalternativeoxidasefamilygnemembers were divided into two subfamilies,namelythe AOX1 subfamily(AkAOX1,AkAOX2,AkAOX3)and the AOX2 subfamily (AkAOX4).GenecolinearityanalysisindicatednopotentialduplicatedgenepairsortandemduplicationeventsinAmopophalus konjac.Ka/Ksanalysisfurthershowedthatthesehomologousgenepairswereprimarilyunderpurifyingselectionduringevolution.The promoteregionsof theAmorpopalusonjacalterativeoxidasefamilygenecontaiedthreetypesofcisactingelements,socated with plant hormoneresponse,growthanddevelopmentresponse,andenvironmental(stress)response.TheqRT-PCRresults showed thattheexpresioleveloftheAkAOXlgeneassignificantlyup-egulatedundersoftrotpathogenifection,droughttress,andsalt stres,whileteexpresionlvelofteAAOX4gneassigficantlydowreglatedundersofrotpathogenifectio.ItwsspeculatedthattheAmorphopalluskonjacalternativeoxidasefamilygeneplayedimportantrolesinitsgrowthanddevelopment,orone signal transduction,and responses to biotic and abiotic stresses.
