Establishment of a rapid propagation system for callus regeneration and seedling formation ofUncaria rhynchophylla

Abstract：UsingthetenderstemsegmentsofUncariarhynchophyllasexperimentalmaterials，suecultureechquesincudingex plantsurfacesterilation，rowigotrolalsidutionnddieretainddentitisdutioneseily studied.Theresults showed thattheoptimal sterilization methodforUncaria rhynchophyllexplants was soaking in 75% ethanol for 30 seconds，rinsingwith sterile water 3 times，followed by treatment with 0.1% HgC 2 solution for8 minutes.The additionof 7mg/L ascorbic acid（VC） effctively reduced explant browning.The optimal medium for callus induction was MS +1.0 mg/L 6-BA+1.5 mg/L 2，4 -D；the optimal differentiation medium was MS +0.2mg/L NAA+2.5 mg/L 6-BA；and the optimal medium for adventitious root inductionwas1/2MS +1.5mg/L NAA.
