Research on optimization of ultrasonic-assisted extraction process of flavonoids from Stephania kwangsiensis H. S.Lo and its antioxidant activity

Abstract:TooptimieeextractionprocesoftalfvonidsfromthetubersofStepaniaangsiesisH.S.Loadexploritsinvitroantioxidantactivity,asedonsinglefactorexperiments,anorthogonaltestmethodwasusedtoinvestigatethefectsofhaol concentration,solid-lqudratioextractiontemperatureandulrasonic imeontheextractionrateoftotalflavooids.Theotalextractionprocesswasetermined,andtheinvitroantioxidantactivityoftotalflvonoidsasfurtherstudied.Theresultsshowedthathe optimalextractionconditionfortotalflavonoidsfromthetubersofStephaniakwangsiensisH.S.Lowasethanolconcentrationof 70% , solid-liquid ratio of 1:35 ( g/mL ),extraction temperatureof 50Y ,and ultrasonic timeof 5O min.Under thiscondition,theyield of total flavonoids content could reach up to 5.45mg/g . The total flavonoids from Stephania Kwangsiensis H.S.Lo exhibited strongantioxidantactivity,with the scavenging rate against 1,1 -diphenyl-2-picrylhydrazyl(DPPH) free radicalsreachingup to (84.39±0.61)% : Inthisstudy,thultrasoundasistedextractionprocesof totalflavooidsoupledwithulravioletspectrophotometryassiplend yieldedreliablersults.Gventhattetotalflavoodspoessedgodantioxidanproperties,thismethodcouldprovideasoundeo reticalreferenceforthe extraction,development,andapplicationof totalflavonoidsfrom Stephania kuangsiensisH.S.Lo. eywords:Stephania kwangsiensis H.S.Lo;totalflavonoids;extractionprocess;orthogonaltest;antioxidantactivity
