Content of 2-Phenylethanol and its glycosides and expression of associated genes in different cherry tomato cultivars

Abstract:Fourcherrytomato(Solanum lyopersicumL.)cultivars(Qianxi,Xiaohong,T7,andFushan88)wereusedasexperiental materials.The contents of 2-phenylethanol and 2-phenylethyl β -D-glucoside in fruits were determined by ultra-performance liquidchromatographycoupled withquadrupole time-of-flighttandemmasspectrometry(UPLC-QTOF-MS/MS).Potentialuridine diphosphate glycosyltransferase(UGT)genes werescreened through phylogeneticanalysis.Theirexpressionpaterns were analyzed by reverse transcriptionquantitative polymerasechainreaction(RT-qPCR)to investigate thecorelation between thecontentsof 2- phenylethanol/ 2⋅ phenylethyl β-D-glucoside andrelated gene expression levels.The results showed significant diferences in 2- (204号 phenylethanol and 2-phenylethyl β -D-glucoside contents among cultivars,with the order of Qianxi>Xiaohong>T7 > Fushan 88.The highest contents of 2-phenylethanol(3 906.69 ng/g )and2-phenylethyl β -D-glucoside(5 590.26 ng/g)were detected in Qianxi, bothsignificantlyhigherthanthoseinothercultivars.Phylogeneticanalysisidentified2OSlUGTgenes.Amongthem,SlUGT42, SUUGT54,SlUGT103,SlUGT104,and SlUGT105showed increasing expresion trends during fruit development.SlUGT103, SUUGT104,andSUGO5exhibitedthehighestexpresionlevelsinQianxi,coincidingwithitspeak2-phenlethanolandglcoside contents.TherelativeexpresionofSUGO3showedsignificantpositivecorrelationswithboth2-phenylethanoland2-pyletyl β-D-glucosidecontets.TherelativeexpressoofSUGO4wassgnificantlypositivelycorelatedwith2-phenylethanolontent. SlUGT103and SlUGT1O4 wereidentifiedaskeyglucosyltransferase genes catalyzingtheglycosylationof2-phenylethanol.
