Full-length transcriptome sequencing and bioinformatics analysis of the Bombyx mandarina

Abstract:Thisstudyemployedsecond-generationsequencing(lluminaRNA-Seq)tocalibrate third-generationsequencing (PacBioISO-Seq)forconductingful-length transcriptomesequencingoftheBombyx mandarina,aimingtoinvestigategnexpres sioncharacteristsduringdiferentpupaldiapauseanddeeplyexploregenomicfunctionalinformatio.Atotalof93616fullength transcripts wereobtainedthroughsequencingandassembly,withasequencelengthof327~3273bp,anaveragelengthof2631bp, andN50of3204bp.IntegrationofannotationsfromCOG,GO,KEGG,KOG,Pfam,SwissProt,eggNOG,andNRdatabasesidentified82796functionallyannotatedgenes.Analysisoffullength transcriptsrevealed17189lncRNAs,87921SSRmarkersand 49432openreading frames(ORFs),withORF-encoded proteinsrangingfromOto1522aminoacids(average305).Gene expressionanalysisacrossdiapausestagesidentified578diferentiallyexpressedgenes,including269withGotationsand1590 with KEGG annotations.
