[Abstract]ObjectiveToinvestigatethemechanismofactionofRadixTrichosanthisinthetreatmentof diabetesmellitus based on network pharmacologyand moleculardocking.MethodsTCMSPdatabase was used toobtain theffectiveconstituents of Radix Trichosanthis,nd SwissTargetPredictiondataplatform wasusedtoobtainthecoresponding targets.MalaCardsdatabase wasusedtoobtainthetargetsasociatedwithdiabetesmelltus.Thecommontargetsofthedrugandthediseasewereobtained,and then STRING database and Cytoscape software wereused toconstruct a protein-proteininteraction(PPI)network andidentifythe coretargets.Matescape database was usedto perform the gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes (KEGG)analyses.Autodock Vina wasusedtoperformmoleculardockingbetweenthecoretargetsandthecorespondingeffective constituents.ResultsAtotalof28targets wereobtainedforRadixTrichosanthisinthetreatmentofdiabetesmelitus,and10 keytargets wereobtained bytopologicalanalysis.TheKEGG pathwayenrichmentanalysisobtainedthesignaling pathways including the peroxisome proliferator-activated receptor(PPAR)signaling pathway.Moleculardocking showed thatthecompounds in Radix Trichosanthis hadgood binding activitytothetargets in thePPAR signaling pathway.ConclusionRadix Trichosanthis may exert a regulatory effect on diabetes mellitus through the PPAR signaling pathway.
