[Abstract]ObjectiveToinvestigate theeffectof polydopamine nanoparticles(PDANPs)onthe motor behaviorof mice with Parkinson’s disease(PD)inducedby6-hydroxydopamine(6-OHDA).MethodsMaleC57BL/6 mice,aged8 weks,were selectedas experimentalanimalsand weredivided intoControl group,PDANPstreatmentgroup(PDA group),modelgroup(6- OHDA group),and combined treatment group(6-OHDA + PDA group). The stereotactic injection technique was used to inject 2 μL of working solution intothecorpus striatum.Aftertwo weeks,the open field test,therotarod test,andthe pole test were usedto assessthe motor behaviorof mice.ResultsCompared with theControl group,the6-OHDA group hada significant reduction in total distance in the open field test (F=14.180,q=8.855,P<0.001) ,significant increases in the time to turn around and the time from the top to the bottom of the pole in the pole test (F=22.090,6.912,q=10.960,5.865,P<0.01) ,and a significant reduction in the latency on the rotarod in the rotarod test (F=8.635,q=6.628,P<0.001) .Compared with the 6-OHDA group,the 6-OHDA+PDA group had a significant increase in total distance in the open field test (q=5.821,P<0.05) ,significant reductions in the time to turn around and the time from the top to the bottom of the pole in the pole test (q=7.232,4.192,P< 0.05),and a significant increase in the latency on the rotarod in the rotarod test ⟨q=4.764,P<0.05) .ConclusionPDA NPs treatmentcan aleviate 6-OHDA-induced motor behavioral damage in mice and improve spontaneous movementand coordination disorders in mice.
