[Abstract]ObjectiveTo prepare glabridin nanoparticle,andto investigateits efectonmotorfunctionin mice with Parkinson'sdisease(PD)induced by1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP).MethodsGlabridinand polydopamine(PDA)wereused tosynthesize glabridin nanoparticles(Glab-PDANPs)through self-assmbly,andthen Glab-PDA NPs werecharacterized.A total of 24 C57BL/6 mice,aged 8 wees,were randomlydivided into MPTPgroup,Glab-PDA NPs treatment group,and control group,with 8 miceineach group.The micein the MPTP group and the Glab-PDA NPstreatment group were given intraperitoneal njectionof MPTPtoestablishamodelofsubacutePD.The miceinthe Glab-PDANPs treatment group were given injectionofGlab-PDANPsthroughthecaudalveinsinceday3before modelingtilltheendof modeling,whiletose in thecontrolgroup wereinjected withanequalvolumeof normal saline.Therotarodtestandthegaittest wereusedtoasessthe motorabityof mice.ResultsThe homogeneous round Glab-PDANPs were prepared succesfull,witha drug encapsulation efficiency of 40% and a drug loading content of 8.7% . The rotarod test showed that the MPTP group had a significantly worse motor coordination ability than the control group (F=5.97,q=2.778,P<0.05) ,while the Glab-PDA NPs treatment group showed significant recovery of motor function . The gait test showed that the MPTP group had significant reductions in the motor ability of the limbs (F=3.03-52.82,q=2.374-9.521,P<0.05) ,while the Glab-PDA NPs treatment group had significant improvements in the indicators of the limbs ⋅q=2.826-8.198,P<0.05) .ConclusionGlab-PDA NPs can improve motor function and restore motor ability in PD mice.
