Effects of acetazolamide on atherosclerotic plaque formation and calcification in miceLIU Han ,AN Yi (Department of Cardiology,The Affiliated Hospital of Qingdao University,Qingdao 266oo3,China)

[Abstract]ObjectiveToinvestigate the efects of acetazolamideonatherosclerotic plaque formationand vascularcalcification in mice. MethodsFour-week-old Apoe-/-(n=3) and wild-type ( n=3 ) C57BL/6 mice were fed for 21 weeks,and subsequently,body weight and serum lipid levels were compared. Twelve l2-week-old Apoe-/- mice were divided into control group ( n=8 ,given normal saline) and experimental group ( ,given acetazolamide at 40mg/kg .After 2 weeks of treatment,aortic tissues wereharvestedtoevaluate plaquearea withHEstaining,characterizelipidcomposition withOilRedOstaining,measure calcium content using calcium assay kits,and determine the expression of carbonic anhydrase I (CA2)by Western blot. Results Apoe-/- mice exhibited significantly higher body weight and serum lipid levels than wild-type mice (F=24.15-61.24,t=2.891- 7.229,P<0.05) . At 3 months of age, plaque area,calcium content,and CA2 expression in aortic tissues were significantly increased in Apoe-/- mice (t=2.872-13.360,P<0.05) .Compared with controls,acetazolamide-treated Apoe-/- mice showed significantly reduced CA2 expresson,lipid-rich necrotic core area,and calcification severity (t=3.820-9.701,P<0.05) .ConclusionAcetazolamide attenuates atherosclerotic plaque formationand caleification by suppressing CA2 expressioninmice.
