[Abstract]ObjectiveToinvestigate thespontaneous activityof substantianigra dopaminergic neurons in twomouse models of chronicParkinson’sdiseaseinducedby1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)andalpha-syucleinprefored fibrils ( α Syn PFFs),respectively.MethodsTwo mouse models of chronic Parkinson’s disease were prepared bylong-term injection of MPTP into the peritoneal cavity and stereotaxic injection of α -SynPFFs into the right striatum of the brain,respectively. Werecorded thespontaneousfiringactivityofdopaminergicneurons inthesubstantianigra parscompactaof the two modelsusing electrophysiology techniques. ResultsThe MPTP group and α Syn PFFs group showed significantly smaller mean numbers of firing dopaminergic neurons per recording session in the substantia nigra pars compacta (t=3.015,4.779 ,both P<0.05 ),significantly higher firing rates of nigral dopaminergic neurons ,both P<0.05 ),and significantly higher proportions of nigral dopaminergic neurons exhibiting burst firing (t=-3.611,-2.928 ,both P<0.05 ),compared to the control group. ConclusionMouse models of chronic Parkinson’s disease induced by MPTP and α Syn PFFs differ significantly from control mice, mainly manifesting withdecreasednumbersofneuronsexhibiting spontaneousfiringandincreasedfiringratesand proportionsof neurons exhibiting burst firing.
