[Abstract] ObjectiveTo investigate the effect of 1-methy-4-phenylpyridinium ( MPP+ )-induced inflammation on the expresionlevelsofDectin-1andinduciblenitricoxidesynthase(iNOS)inBV2microglialcels.MethodsBV2microglialcels were treated with different concentrations of MPP+(0.1,0.5 ,and 1.0mmol/L ,and MTT assay was used to measure cell viability. Western blotting was used to observe the effect of different durations of MPP+ treatment(2,4,6,12,and 24 hours)on the proteinexpressionlevelsofthepaternrecognitionreceptorDectin-1andtheinflammatoryfactoriNOSinBV2microglialcels. Results MPP+ at the concentration of 0.5mmol/L and 1.0mmol/L significantly reduced the viability of BV2 microglial cells ( (F=9.058;q=3.611,4.361;P<0.05) . There was a significant increase in the protein expression level of Dectin-1 at 4,6,and 12 hours after MPP+ induction (F=5.660,q=4.784-6.610,P<0.05) ,which returned to normal at 24 hours. There was a significant increase in the protein expression level of iNOS at 12 and 24 hours after MPP+ treatment (F=4.884;q=3.662,3.931;P< 0.05).Conclusion MPP+ -induced inflammatory response in BV2 microglial cells can significantly increase the protein expression of the pattern recognition receptor Dectin-1 and iNOS.
