[Abstract]ObjetiveToinvestigate theprotective efectof demethyleneberberine(DMB)against mitochondrial injuryin duced by 1-methyl-4-phenylpyridinium ( MPP+ )in SH-SY5Y cells.MethodsSH-SY5Y cells were divided into control group, (204号 MPP+ group,selegiline+MPP+ group,and DMB+MPP+ group. CCK8 assay was used to observe the effect of different concentrationsofDMBonthesurvivalrateofSH-SY5Ycels,andflowcytometrywasused to measure mitochondrialmembranepotential (2号 (Δψm) and reactive oxygen species (ROS).ResultsThere was a reduction in the survival rate of SH-SY5Y cels after treatment with 1 mmol/L MPP+ for 24 hours,and different concentrations of DMB could improve the survival rate of SH-SY5Y cells, among which the co-treatment group with 10-3μmol/L DMB and MPP+ had the highest survival rate ⟨F=74.5,q=18.12,P< 0.001). MPP+ treatment reduced Δψm in SH-SY5Y cells ⋅F=185.2,q=33.08,P<0.001) ,while both DMB and selegiline could reverse the effect of MPP+(q=12.97,15.32,P<0.001) 1 MPP+ treatment increased the content of ROS in cells ( ⋅F=79.2,q= : ?0.88,P<0.001 ),while DMB and selegiline eliminated the effect of MPP+(q=14.62,15.08,P<0.001) .Conclusion DMB can alleviate mitochondrial injury in SH-SY5Y cells induced by MPP+ :
