Effects of extraction process and source of Auricularia heimuer melanin on its UV-Vis spectrum and antioxidant activity

Abstract:Auriculariaheimuerisatraditionaledibleand medicinal fungus,anditsmelaninexhibitsdiverse biological activities,includingantioxidant,antitumor,andimmunomodulatoryproperties.Topromotetheeficientdevelopentand utilization of A .heimuer resources,this study systematically evaluated melanin extracted from different sources and by variousmethodsbasedonyield,UV-Visspectralcharacteristics,andinvitroantioxidantactivity.Theresultsdemostrated thatenzymatichydrolysis Was theoptimal methodforextracting melanin fromfruitingbodies,achievingthehighestyield ( 9.49% )and exhibiting the strongest antioxidant capacity.At a concentration of 2.0mg⋅mL⋅1 ,the melanin scavenged DPPH,ABTS, and O2- with efficienciesof 80.08% 82.87% ,and 63.47% ,respectively. In contrast,melanin derived from liquid fermentation broth exhibited superior scavenging activity against DPPH and O2-⋅ ,when the sample concentration was 2.0mg⋅mL-1 ,the scavenging rates were 98.52% and 80.61% ,respectively,and displayed UV-Vis spectral features mostsimilartothoseof the standard melaninreference.Thesefindings provideasolidtheoreticalfoundationforoptimizing the extraction ofA. heimuer melanin and advancing its application in functional product development.
