Abstract:Tostudytheeffctsofdifferent preservation methodsonthe traitsofAuricularia heimuerstrains,1Ostraisof A.heimuer that had been preserved for2O years by liquid nitrogen preservation method and 4∘C slant transplantation methodrespectively were used as thetest materials.Methods such as determinationof growth status,cultivationfor fruiting bodyproduction,esterase isoenzyme analysis,and ISSR molecularmarkeranalysis wereadopted to investigate the long-term preservationeffectsofdiferent preservation methods.Theresults showed thatfor thesame strain,in termsof mycelialgrowthvigorandgrowthrate,theoverallperformanceof liquid nitrogen preservation was significantlybetter than that of the 4∘C slant transplantation method.There was no significant diffrence in the aspect of fruiting body production.For thesame strainunder different preservation methods,theclusteringanalysisof the esterase isoenzyme indicated wasuniformlyl,andthesimilaritycoefficients inthe ISSRmolecular markerclusteringanalysis Wereall higher thanO.90.Thisindicates that thesamestrainunderdiferentpreservationmethodsishighlysimilarbuttherearestillcertain diferences.It is likely that changes occurred during the long-term preservation of the strain by the 4∘C slant transplantationmethodor during thesubculture process.This studyisof great significanceforthe long-term preservationof ΠM heimuer strains and maintaining stability during the process of industrial production.
