Abstract:Toexplorekeygenes involved inLentinulaedodes(shitakemushroom)responsetoheatstress,transcriptome sequencing analysis was performed using the heat-tolerant strain Xiang 931and the heat-sensitivestrain Nongke1.Myceliawere subjected to high-temperature treatment at 36∘C ,with normal temperature cultivation serving as the control (CK).The results showed that compared to the CK group,the mycelial growth rate of both Xiang 931 and Nongke l significantlydecreasedafterhigh-temperature treatment.Strain-specificdifferentiallyexpressed genes(DEGs)inresponse to high temperature were740for Xiang 931and 702 forNongke1.KEGGand GOenrichment analysis of the DEGs revealedthat genes fromboth strains wereprimarilyandsignificantlyenrichedinmetabolism-relatedfunctional pathways and processes,although the number of enriched genes differed between the two strains.Combinedanalysis based on KOG anotation and relevant databases indicated that laccase genes and HSP2O family genes exhibited significant expression differences between treatments,suggesting theyare potential regulatory genes associated with heattolerance in L edodes. Six DEGs wererandomlyselectedforqRT-PCRvalidation,andtheresultsconfirmedthatthegene expression trends were consistentwiththetranscriptomesequencingdata.Thisstudyprovidesafoundation forresearchintothemolecularmechanismsofheat tolerance inL.edodesand the breedingof newheat-tolerantcultivars.
