Abstract:Todevelopmolecular markersfor Cucurbitamaxima rootstocksrelated to theirabilitytoremovecucumber fruitsurface'sbloom,evaluated18different C. maxima materials with varying bloom removal capabilities through grafting.Theresults showed that whencucumberwasgraftedonto rotstocks with strong bloomremovalabilities,the silicon content inthestems,leaves,and fruitsof the scioncucumber was significantlylower compared to those grafted ontoootstockswith weak bloom removal abilities.This indicates that the bloom removal capability of C maximaisassociated with its ability to absorband transportsilicon. Using the previouslyreported molecular marker CM1-CAPSbased on SNP of the silicon transporter gene CmLSiI ,alongwith the SNP-Sanger sequencing and Hi-Tom sequencing methods designed in this study,we identified and analyzed 57C maxima rootstockswith varying abilities to bloom removing. The results showed that all three identification methods achieved a 100% accuracy rate for selecting the bloom removing trait, indicatingthat this molecular markeris suitableforbreeding bloomremoving rootstocks,which isof greatsignificance toaccelerate the breeding of newvarieties.
