Abstract:Theinibitoryefectofmonoclonalantibodyagainstearlypregnancyfactor(EPF-B6)onthebiologicalbehaviorofcervical cancer HeLacellswasinvestigatedbystudyingtheeectsofEPF-B6ontheproliferation,migration,cloningandapoptosisofHeLa cells.RT-PCR and Western Blot were used to detect the expression of EPF gene in six common cancer cells and EPF protein in cell culture medium.TheeffctsofdiferentconcenrationsofEPF-B6monolonalantibodyontheproliferation,migrationndpopulation dependenceofHeLacelsweredetectedbyCCK8assaycellscrathassayandplatecloning assay,espectively.Theapoptosisand cycleofHeLacellswere detected by flowcytometry.Results showed that EPF gene was present in common tumor cells.EPF protein was expresed in theculture medium ofcervicalcancer HeLacells,breastcancer MCF-7celsandovariancancer A2780cels.The resultsshowedthatEPF-B6monoclonalantibodycouldsignificantlyinhibit theproliferation,migrationandcolonyformationofHeLa cells,it was concentration-dependent ( P<0.01 ).The results of cell cycle analysis showed that EPF-B6 monoclonal antibody blocked the DNA replication process of HeLa cells at G2/M phase.
