Preparation of polyclonal antibody against coat protein of pinellia isolate of soybean mosaic virus

Abstract：Toestablish arapid detection methodforsoybean mosaic virus（SMV），the primary pathogencausing mosaic disease in Pi neliaternata，thecoatproteingeneofSMVwasamplifiedbyreversetranscription-polymerasechainreaction（RT-PCR），anditsro karyoticexpressionvectorwasconstructed.Thepurifiedfusion-expressd proteinwasusedasanantigen toimmuizerabbitsforthe preparation of polyclonal antibodies.The results showed that the prepared antiserum exhibited a titer of 1：102，400 as determined by enzyme-linked immunosorbent assay（ELISA），a Western blot detection limit of 300ng for the antigen，and demonstrated strong specificity.
