Preparation of polyclonal antibody against coat protein of pinellia isolate of soybean mosaic virus

Abstract:Toestablish arapid detection methodforsoybean mosaic virus(SMV),the primary pathogencausing mosaic disease in Pi neliaternata,thecoatproteingeneofSMVwasamplifiedbyreversetranscription-polymerasechainreaction(RT-PCR),anditsro karyoticexpressionvectorwasconstructed.Thepurifiedfusion-expressd proteinwasusedasanantigen toimmuizerabbitsforthe preparation of polyclonal antibodies.The results showed that the prepared antiserum exhibited a titer of 1:102,400 as determined by enzyme-linked immunosorbent assay(ELISA),a Western blot detection limit of 300ng for the antigen,and demonstrated strong specificity.
