Establishment and evaluation a high performance liquid chromatography method for the determination four aflatoxins in wheat flour

Abstract To establish an efficient method for the determination aflatoxins B1 , B2 , G1 ,and G2 (AFB 11 ,AFB2,AFG1, AFG 2 ) in wheat flour,this study systematicallyoptimized sample pretreatment and chromatographic conditions basedon high-performance liquid chromatography with fluorescencedetection (HPLC-FLD).Theresults showed thatacetonitrile一 water(80:2O,v/v)combinedwith sodium chloride salting-out as the extraction solvent significantly improved the recovery targetanalytes.After purification with immunoafinitycolumns and derivatization with trifluoroacetic acid, matrix interferences were effectively removed.Under the optimized chromatographic conditions,the four aflatoxins exhibited good linearity in the concentration range 0.1-20.0ng/mL ,with correlation coefficients (r2) all greater than 0.999 9.Method validation results indicated that the limits detection (LOD)ranged from 0.008 to 0.017ng/g ,and the limits quantification (LOQ)were 0.026-0.055ng/g .Atspiked levels 0.5-5.0ng/mL ,theaverage recoverieswere 80.2%-89.5% ,with relative standard deviations (RSDs) all less than 2.0% . The RSD the six-replicate stability test was below 1.5% , and the RSD for quality control sample analysis was less than 0.12% , demonstrating good method stability.Inconclusion,thedeveloped method featuredsimple pretreatment,highsensitivity,andsatisfactoryaccuracy, and was suitable for the trace determination multiple aflatoxins in wheat flour.
