Root rot pathogens of melon and their pathogenicity in Kashi, Xinjiang

Abstract:Inorder toclarifythepathogenspeciesand theirpathogenicityofmelonrootrotinGashicountyKashgar region,Xinjiang.Samplesof melonrootrotwerecollcted,pathogen isolatedandidentification.Thepathogenswere isolatedandpurifiedbytissue isolation method,andverificatedbyKoch'slaw.Thenpathogens wereidentificatedbycombiningmorphological characteristicsand phylogenetic analysis based onmulti-loci(ITS + TEF1- a+ RPB2).Theresults showedthatthe pathogens of melonrootrot in Gashi county weremainlyNeocosmospora falciformis,Fusarium glycines and N. solani.Among them,strains Z3,Z11 and Z27,withhigh pathogenicity,were inoculatedon thesame melon variety todetermine the pathogenicity.The results demonstrated thatthese three strains significantlyreduced theroot length, plantheight,stem massandrootmassofmelonseedlings,withthedisease indexesof68,84and74forZ3,Z1land Z27, respectively. Strain Z11 (F. glycines)had the strongest pathogenicity. In this study, N. falciformisand F. glycines were identifiedforthefirsttimeaspathogensofmelonrootrotinXinjiang,whichenrichedtheetiologyofmelonroototand provided a theoretical basis for the diagnosis,control and resistance breeding of melon root rot.
