YUJiang-tao,WUXu-ting,HUDa-guietal(CollgeofHorticulture,NanjngAgricultural University,Nanjing,Jiangsu2095) Abstract [Objective]Arapid propagationsystem for“Suzhen No.1”wasestablished through tisueculture techniquesandleaf-induced regeneration,providingatechcalfoundationforislargescaleadficientmiplicatioMetod]Domantudsof“SuenNo”wee usedasexplantsforpriarytisueculture,andtheirgrowthstatuswerecompaedtoconfimtheostsuitableminimalmedium.Teoptial subcultureconditioswereidentfiedytestingcombinatiosofIAAandatvaringconcetratios.eotialhoronconditioforthe regenerationofleaveswereconfiedbycombingdiferentconcentrationsofDZandNAA.Result](/2N)MSwasidentfiedaseotimal minimal medium for“Suzhen No.1"tissue culture. The dormant bud germination rate reached 76. 0% ,with vigorous plant growth and tender green leaves;subcultured shoots exhibited the best growth on(1/2N)MS + IAA 0.1 mg/L + ZT1.O mg/L,reaching an average height of 37.8mmafter3,withaproliferationcoefcietof4.6,igrconentrationofZ3.Omg/Lexeredanibioryeetoneowthf subcultured shoots;Leaf explants showed optimal regeneration on (1/2N)MS + TDZ 3.0 mg/L + NAA 0.2 mg/L,achieving acallus induction rate of 90.0% and an average of3.1adventitious shots per explant.[Conclusion]Arapid tisse culture propagation systemfor“Suzhen No. 1"was established: (1/2N)MS medium was the most suitable basal medium; (1/2N)MS supplemented with 0.1 mg/L IAA and 1.0mg/L (2号 ZT provided optimal conditions for subculture;(1/2N) MS supplemented with 3.0mg/L TDZ and 0.2mg/L NAA was optimal for leaf callus induction.
