Screening, identification, and growth- promoting effects on tomato of high-yield IAA-producing Bacillus
Abstract:Toscreen Bacilusstrainwithhigh Indole-3-aceticacid(IAA)production from therhizosphereofsweetcherries, clarifytheirgrowth-promotingcharacteristicsuchassiderooresecretion,phosphatesolubilation,potassiumsolbilization,andnitrogen fixationas wellas their growth-promoting efects,soas to provideabasis forthesubsequent developmentof microbial fertilizers forsweetcherry.Spectrophotometry wasusedtodeterminetheIAAproductionofplantrhizospheregrowth-promoting bacteria(PGPR).Strain identification was conducted throughmorphologicaland16SrRNA sequence analysis.Qualitative detection wasperformedonthe growth-promotingcharacteristicsofthe strains including phosphate solubilization,potassium solubilization,andnitrogen fixation.Through the germination experimentof themodel plant,tomato,the growth-promoting efectofstrainsonplants was verified.Theresultsshowed thatfourhigh IAA-producingBacillus strainswere screened out,with IAA production ranging from14.79 to 20.71mg⋅L-1 .Theywere identified as Bacilusvelezesis(A81),aciluspaiicus(E2),Bacillusznthoxyli(15),ndcillsbus(R42),espeivly growth diameter of all strains ranged from 0.61 to 0.77cm ,and all had the abilities of acid production,siderophore secretion,organicphosphatesolubilization,potassiumsolubilization,andnitrogenfixation.Allstrainshadsigniicant growth-promoting effects on tomato seedlings,with the growth-promoting effects in the order of R42>S151> A81 > E2. Compared with thecontrol(CK),R42and S151 significantly increased the plant height by 26.39% and 20.00% ,respectively,while their root length significantlyincreased by 17.51% and 16.61% ,respectively.In conclusion,the rhizosphere of sweet cherryharbors multifunctional growth-promotingBacillus with highIAA production,and theresultsof this study provide important bacterial resources for the development and applicationof functional microbial fertilizers.
