Abstract:GenomicDNAextracteddirectlyfromplantseedscouldbeusedformolecularidentificationofseedpurityand specific genotypes.There is no report on effcientdirect extraction methods for genomic DNA from single drycelery sed untilnow.Inthisstudy,thesingledriedceleryseed wassubjectedtoextractgenomeDNAbyalkaline lysismethod, which wasutilizedasthe template forPCR(polymerase chainreaction)amplification withActin-specific primers to verify whether theextractedgenomicDNAmets therequirements formoleculardetection.Theresultsshowedthatclearspecific bands wereobtained byPCRamplification,demonstratingthatthegenomic DNAdirectly extracted fromsingle drycelery seed was suitable for PCR amplification.To assess DNAstability,PCR amplification was conductedusing the genomic DNA stored at 4∘C for14 days,and successful amplification was confirmed by the presence of expected bands in agarose gels.Thisstudyestablishedarapid,simpleandhighlyefficient methodforextractinggenomicDNAfromindividualdried celeryseed.Theextracted genome DNAwassuitableforPCRamplificationand exhibited good stability.Itcouldbeused forMAS ofcelery seed purity and specific genotypes.
