Abstract:To investigateDNAextractionmethods frommelon leavessuitableforKompetitiveAlele-Specific PCR (KASP)marker detection system,this study used melon parents B551,B22 and their F1 hybrid B551×B22 )asmaterials.Three DNAextractionmethods includingalkaline lysis,CTABandacommercial magnetic bead-based kit-were employed to extract DNA from melon leaves.The efects ofthese three DNA extraction mehods on the KASP marker detectionsystem wereanalyzed.DNAqualityassessmentresultsshowed thatthecommercial kit method efectivelyremoved impurities such as proteins,polyphenols,and polysaccarides from melonleaves,achievingaKASPdetectionaccuracyof 100% .TheCTABmethodachievedadetectionaccuracyof 97.2% ,whilethealkalinelysismethodachievedadetectionof 69.4% . In conclusion,thecommercial magnetic bead-based kitis the mostsuitable for high-throughput KASPdetection, the CTAB methodcanbeused forsmal-scaledetection,andthealkalinelysis methodisnotsuitableforDNA extraction from melon leaves in KASP marker detection systems.
