Functional analysis of the StWRKY70 gene promoter in potato

Abstract:The transcription factor StWRKY7Oisinvolved inplant diseaseresistancedefenseresponsesandcanenhance potato early blight resistance.To investigate the promoter function of the potato StWRKY70 gene promoter,this study clonedthe promotersequenceusingPCR methodand predictedits cis-acting elements usingonlinetoanalyzethedistributioncharacteristicsofregulatoryelements.Aditionall,5'-enddeletion expression vectorofthepromoter(pGreenI 0800-StWRKY7Opro:LUC)were constructed.The expression of thereporter gene LUC wasobserved through transient transformation of tobacco leaves to identifythe promoter’sactiveregions.Theresults showed that the length of the promoter sequencewas2ooo bp,andcontainedvarious stres-responsivecis-elements,suchasdehydration-responsiveelementMYB,drought-responsiveelementAP2/ERF,andhigh-temperature-responsive elementHSF.Theanalysis resultsof LUCfluorescence intensity showed that with the increase of the length of different 5′ -end deletion fragmentsof the St WRKY70 gene promoter,the LUC fluorescence intensitygradually increased,indicating thatthecoreregionof the St. WRKY70promoterwas 400bpto70o bp.Inconclusion,thecoreregionandvarious stress-responsive cis-acting elements ofthe StWRKY70 gene promoter may be involved in the potato's defense response against early blight byregulating gene transcriptioneficiency,therebyprovidingcertaintheoreticalsupportforfurtherelucidatingthemechanismunderlying the role of StWRKY70 in plant disease resistance.
