Study on the Antioxidant and Hypoglycemic Abilities of Crocetin in Gardenia jasminoides Ellis

Abstract: Objective: To investigate the antioxidant and hypoglycemic effects of crocetin in Gardenia jasminoides Elis.Method: The scavenging abilityof crocetin on ABTS+freeradicals and its inhibitory activities on (20 a -amylase and a -glucosidase were evaluated through in vitro antioxidant enzyme and digestive enzyme inhibition tests,and its hypoglycemic effect was investigated using the insulin-resistant HepG2 cell model.Result:The half maximal inhibitory concentrations (IC50) of saffron acid on ABTS+ free radicals, a -amylase, and αa -glucosidase were 16.70,35.23μg⋅mL-1 and 7.58μg⋅mL-1 ,respectively, showing a significant dose-dependent inhibitory effect; in cell experiments,crocetin significantly promoted glucose uptake in insulin resistant HepG2 cels. Conclusion: Crocetin derived from Gardenia jasminoides Elis hasantioxidantcapacityand hypoglycemic activity,which provides important theoretical basis for the development of functional foods for preventing metabolic diseases.
