Abstract:WRKY transcription factorsplayacrucialroleinthesignaltransductionpathwaysofplant diseaseresistance. Potato was selectedasthe modelorganism to investigatethe propertiesand functions of StWRKY15.RNA-seqanalysis identifiedone transcriptionfactor,StWRKY15,asbeingsignificantlyinducedbypotatoearlyblight.StWRKY15wassuccessfullyclonedusingPCRtechniques.Theful-lengthsequencespans1O02bpandencodes33aminoacids.Subcelular localisation was confirmed to bewithin the nucleus.StWRKY15 belongs totheunstable hydrophilic protein family.It has a molecular mass of approximately 33.6kDa andatheoretical pI of around 9.45.It exhibits the highest homology with CaWRKY15.Both proteinsaremembersof the WRKYfamily.AnalysisofStWRKY15expressionlevels invarious potato tisues usingqRT-PCRrevealed thehighest expression instems.The expression levelsof StWRKY15 were measuredat varioustimepointsafter inoculationwithAlternariasolani.TheresultsindicatedasignificantincreaseinStWRKY15expressionat24 hours post-inoculation,with peak expresionbservedat48 hours.This demonstrates that StWRKY15is stronglyinducedbypotatoearlyblight.TransientoverexpressionestablishedthatStWRKY15isassociatedwithenhanced resistance to Alternaria solani in tobacco.These results lay the groundwork for further research into the roleof St. WRKY15 inconferring resistance to early blight in potato.
