Abstract:To investigate the physiologicalregulatory mechanismofsalt stressonpumpkin growthand development,two pumpkinlines(360-3and20-8)with significantlydifferent salt tolerance wereusedasexperimental materials.A seedling-stage salt stress experiment was conducted tosystematicallyanalyze their physiological response characteristics. ThreeNaCl treatment concentrations of O, 20o,and 300mmol⋅L-1 were applied to examine the effects of salt stress on seedling growth,physiologicalmetabolism,andantioxidantenzymeactivity.Theresultsshowedthatwiththeincreaseof NaCl concentration,theseedling heightofboth lines decreased significantly,withagreater reductionobserved in20-8. The relative watercontent(RWC)of leaves anddry freshmass decreased with the increaseof saltconcentration; However, RWC inthesalt-tolerant line 36O-3remainedstable and was significantly beterthan thatofO-8.Salt stressaggravated cell membranedamage,andtherelativeelectricalconductivityof thetwopumpkinmaterials increased significantlywith the increase of salt concentration.In the response to antioxidant enzyme activity,under the treatment of 300mmol⋅L-1Na. Cl,the activity of superoxide dismutase,peroxidase,and catalase in 360-3 were significantly increased by 230.91% , 284.72% ,and 40.82% ,respectively, compared with the control,which were substantially higher than 103.03% 24.96% , and 22.89% of20-8,respectively.The hydrogen peroxide content of the two materialsunder NaCl increased by 37.07% and 39.95% ,respectively,indicating that salt-tolerant line 36o-3 more effectively alleviates oxidative damagethroughitsantioxidantsystem.Inconclusion,line36O-3exhibitsstrongersalttolerancethan20-8,mainlydueto its superior water retention capacity and enhanced antioxidant enzyme activity under salt stress.
