AbstractThisstudy utilized transcriptome sequencing to analyze floral tisses of Dendrobium nobileacross 3 developmentalstagesearlybudstage(stageI),latebudstage(stageII),initialfloweringstage(stage)anduder stress conditions (low temperature,high temperature,and mechanical damage,all applied to stageI samples). Comparative analyses of transcriptome profiles were conducted between developmental stages (I vs Ivs Il ,Ivs III) and between stressed samples and their corrsponding controls (stage II stress vs control).The resultsrevealed 26 598,29 680,30 263,7495,13 713,and 8 764 differentially expressed genes (DEGs) for the I vs vs Il ,Ivs ⅢI ,low temperature,high temperature,and mechanical damage comparisons,respectively.Gene Ontology (GO) enrichment analysis indicated that these DEGs were primarily associated with the biological process,cellular component,and molecular functioncategories.Kyoto Encyclopedia of Genes and Genomes (KEGG)pathwayanalysis showed significant enrichment in processessuchas planthormone signaltransduction,aminosugarand nucleotide sugar metabolism,plant-pathogen interaction,and the plant MAPK signaling pathway.Furthermore,74 MADS transcription factors wereidentifiedasdiffrentiallyexpressedacrossthecomparisons,with40,34,40,17,25,and16foundintheI vs ,Ivs II ,Ivs II ,low temperature, high temperature,and mechanical damage comparisons, respectively. These findings provide valuable insights intothe molecular mechansms underlyingfloral development and stressresponses in Dendrobiumnobile.
