Abstract:Pancreaticcanerisahighlymalignanttumorofthedigestivesystem,withasignificantlylowerfive-yearsurvivarate thanothermalignancies.Gemcitabine(GEM)istheprimarychemotherapeuticagentforpancreaticcancer,butitseficacyisoften limitedbychemotherapyresistanceof tumor.Thisarticle elaboratesonthe intrinsiccellar mechanismandnon-cel-autonomous mechanism of GEMresistancein pancreaticcancer and summarizes howto enhance thesensitivityof pancreaticcancercells to GEMbyinducing feroptosisthroughtheregulationofpolyunsaturatedfattacidperoxidation,ironmetabolismcontrol,and antioxidantsystems,inorder toinvestigate theassociationbetweeferoptosisandGEMresistance mechanismsandprovidenew directions for the clinical treatment of pancreatic cancer.
