Phylogenetic Study of Tilia Plant Based on DNA Barcoding

JI Xiao-jing,JHong-taoLUAZhi-uietal(CoegeofLifeiences,hangcunNoalUniversityhangchun,lin01) AbstractTheexperimentadoptedDAbarcodetechnolgytoconductphylgeneticresearchonTiliaplantconstructedapylogetictr ofthegenus Tilia.Theresultsshowedthatthegenesequencelengthobtaindbyprimeramplficationofsample1,Tiliamurensis,was ,with A(adenine)content of 18.96% ,T(thymine)content of 19.41% ,C(cytosine) content of 32.51% ,and G(guanine) content of 29.12% ,with an average A+T content of 38.37% and an average G+C content of 61. 63% : A+T is much lower than G+C ;the length of the gene sequence obtained by primer amplification for sample 2,Tilia mandshurica,was 770bp ,with A content of 42.99% ,T content of (20 30.26% ,C content of 13.51% ,and G content of 13.25% . The average A+T content was 73.25% ,and the average G+C content was 26.75% ,with A+T being much higher than G+C ;the gene sequence length of sample 3, Tilia miqueliana, obtained by primer amplification was 810bp ,with A content of 21. 48% ,T content of 20.00% ,C content of 29.63% ,and G content of 28.89% . The average A+T content was 41.48% ,and the average G+C content was 58.52% : A+T was lower than G+C . Gene sequence analysis was conducted using dnastar 7.Osoftware,dtreepygenetictrsereostructedteteinethegeneticelatioshipetweteexperimetalamplesdplants intheTilapiaceae.TeAbarcodetechnolgynotonlyprovidesconclusiveevidenceforclarifyingthepylogeneticelatioshipsbetwen plants,but also provides theoretical support for efcient analytical frameworks for other species.
