[Abstract] Objective： To analyze the expressions of cytokeratin 5/6 （CK5/6） and P63 protein （P63） in cancer tissuesand their relationship with clinicopathological characteristics in patients with non-smal cell lung cancer （NSCLC）.Method： A retrospective collection was performedon the clinical data of 18O patients with NSCLC in the First People's Hospital of Fuzhou from January 2O22 to December 2023.The expressions of CK5/6 and P63 in cancer tissues and para-carcinoma tissues was detected by immunohistochemistry，and their relationship with clinicopathological characteristics of NSCLC patients was analyzed.Result：The positive expression rates of CK5/6 and P63 in cancer tissues of NSCLC patients were 73.33% and 77.22% respectively， which were higher than 62.78% and 66.67% in para-carcinoma tissues （ P <0.05）. The positive expression rates of CK5/6 and P63 in cancer tissues of patients with TNM stage II-IV，low-moderate differentiation，and squamous cell carcinoma were higher than those of patients with stage I-II ，well-differentiated，and adenocarcinoma， the differenceswere statistically significant （P<0.05） .Conclusion：The expressionlevels of CK5/6 and P63 in cancer tissues are closelyrelated tosquamous cell carcinoma，stage Il-IVand low-moderate differentiation in NSCLC patients，high expressions canbe applied as thebasis to determine pathologicalclasifications，differentiation degree and clinical staging of NSCLC.
