Abstract:Asthefourth largestglobalfoodcrop,potatoholdssignificantimportanceinensuring foodsecurityand promoting agriculturalidustrializatioTradioaltuberpropgationfacschallngessuchaslongycles,loencandsuscetiblito pestanddiseasetransmisio.Inontrast,theinvitrorapidpropagationtechnologyenablestherapidmuliplicationandlargscale productionofvirus-freesedlingsthroughtisueculture.TopreservethequalityoftheWenling Xiaoyangshupotatothisstudy utilizedWenlingXiaoyangshuasexperimentalmaterialtoinvesigatesterilizationmethods,dferentiationmediumfoulatiosnd subculture medium formulations.Theresultsdemonstrated that the optimal sterilizationprotocol involved 75% ethanol for 30 s combined with 0.1% HgC 12 for 7 min,achieving the lowest contamination rate of 1.67% for explants.The optimized conditions for stem tip callus induction were MS medium supplemented with 6.5g⋅L-1 agar, 30g∙L-1 sucrose, 0.3 mg·L -1 NAA (Naphthaleneacetic acid),and 1.5 mg ·L -1 6 -BA (6-benzylaminopurine),yielding the highest induction rate of 91.67% .For subculture,the optimal medium was MS+6.5 g·L -1 agar+ 30g 山 -1 sucrose+0.1 mg·L -1 NAA+0.1 mg·L -1 6-BA.
