[Abstract]ObjectiveToinvestigate thediferentiallyexpressed genes（DEGs）betweencoloncancertissueand normal intestinal tisue using the GEOand TCGAdatabases，toidentifysurvival-related genes，andtoestablishaprognostic predictionmo del.MethodsAunivariateCoxregresionanalysisbasedontheTCGAdatabasewasused toidentifysurvival-related DEGs，and theKaplan-Meiersurvivalcurvewasusedtoanalyzetheimpactof thesegenesontheprognosisofcoloncancerpatients.AmultivariateCoxregressonanalysis wasfurtherconductedtoestablishaprognosticprediction model.Geneontologyfunctionalenrichment analysis andGene SetEnrichmentAnalysis（GSEA）were performedfortheDEGsobtained，andtheasociationbetween tumor mutationcharacteristicsandthelevelofriskwasexplored.Inaddition，awaterfallplotwas generatedforgene mutations.Results TenDEGssignificantlyassociated withthesurvivalofpatientswithcoloncancerwereotainedsuccessfull，ndaprognosticpre diction model was establishedbasedonthese genes.TheresultsofGSEAshowed thattheprognosis ofcoloncancer patients might beasociated withthe signaling pathwayssuch as E2F_TARGETSand MYC_TARGETS.There were significant diferences inthe preferenceforamplification mutations of specific genesand mutational spectra between diferent risk groups.ConclusionThis studysucesfullybtainste genesasociated withthesurvivalofcoloncancerandestablishsaprognosticpredictionmodel.The comprehensiveutilizationofbioinformatics tolsand methodscan providenewcluesandatheoreticalbasisforthepreventionand treatment of colon cancer.
